Making ATP stocks (takes ~15 minutes)

To make 0.1M ATP aliquots:

 

1)      Weigh out ~11g ATP

2)      Dissolve in ~150ml cold water in a 250mL beaker w/stir bar.

3)      Add solid KOH to pH to 7.0.  This is solution A.

4)      Dilute 100uL of solution A into 1 litre of water, and MIX WELL

5)      Take spec reading of this 1:10000 dilution at 260nm blanking against water (OD)

6)      Pour solution A into a 250mL measuring cylinder.  Measure volume. (V)

7)      Calculate final volume (Final volume = V*OD/0.154) and make up to final volume in measuring cylinder with cold water.

8)      Mix well and pour into falcon tubes in 10ml alequats and freeze at -20ēC.
